  • Targeting FcRH5 in MM: Rationale and emerging dual-targeting strategies
    2026/10/12

The Multiple Myeloma Hub was pleased to speak with Adam Cohen, University of Pennsylvania, Philadelphia, US. We asked about the rationale for targeting FcRH5 in multiple myeloma and emerging dual-targeting strategies.

In this interview, Cohen discusses Fc receptor-homolog 5 (FcRH5) as a therapeutic target in multiple myeloma (MM) and its potential role following antigen loss associated with B-cell maturation antigen (BCMA)- or G protein-coupled receptor class C group 5 member D (GPRC5D)-directed therapy. Cohen outlines the rationale for targeting multiple antigens, the potential benefits and challenges of dual-targeting, and which patients may benefit most from these approaches. Cohen also reviews emerging data on the FcRH5-targeted bispecific antibody cevostamab, including its evaluation as monotherapy and as part of combination and sequential treatment strategies.

  • IMS 2026 | CARTITUDE-2 Cohort A: ≥5-year remission and survival with cilta-cel in RRMM
    2026/10/08

During the 23rd International Myeloma Society (IMS) Annual Meeting and Exposition, September 23–26, 2026, Glasgow, UK, the Multiple Myeloma Hub spoke with María-Victoria Mateos, University Hospital of Salamanca, ES. We asked about the long-term outcomes with ciltacabtagene autoleucel (cilta-cel) in patients with relapsed/refractory multiple myeloma (RRMM) in Cohort A of the CARTITUDE-2 trial.

In this interview, Mateos discusses the ≥5-year findings from Cohort A of the phase II CARTITUDE-2 trial (NCT04133636), which evaluated a single cilta-cel infusion in patients with RRMM after 1–3 prior lines of therapy who were exposed to a proteasome inhibitor and are lenalidomide-refractory. Mateos reviews long-term remission and survival outcomes, characteristics associated with prolonged disease control, immune correlates of durable response, and long-term safety findings. Mateos also discusses these findings in the context of cilta-cel use in earlier lines of therapy.

  • IMS 2026 | UKMRA ProMMise D interim results: VRAT-guided belantamab mafodotin dosing in RRMM
    2026/10/06

During the 23rd International Myeloma Society (IMS) Annual Meeting and Exposition, September 23–26, 2026, Glasgow, UK, the Multiple Myeloma Hub spoke with Rakesh Popat, University College Hospital, London, UK. We asked about interim results from Arm D of the UKMRA ProMMise trial in patients with relapsed/refractory multiple myeloma (RRMM).

In this interview, Popat discusses interim results from Arm D of the ProMMise trial, which is evaluating a vision-related anamnestic tool (VRAT) to assess ocular symptoms and function, and guide belantamab mafodotin dosing in patients with RRMM receiving belantamab mafodotin + bortezomib + dexamethasone (BVd). Popat reviews interim results from Stage 1, including the correlation between VRAT and ocular assessments, and outlines the planned evaluation of VRAT-guided dosing without mandated ocular examinations in Stage 2.

  • How to optimize outpatient delivery of bispecific antibody therapy for MM
    2026/09/29

The Multiple Myeloma Hub spoke with Rakesh Popat, UCL Hospitals NHS Foundation Trust, London, UK. We asked how to optimize outpatient delivery of bispecific antibody therapy for multiple myeloma (MM).

In this interview, Popat discusses practical strategies to support the outpatient delivery of bispecific antibody therapy, including patient selection, caregiver support, and access to timely medical care if needed. Popat also outlines approaches that could facilitate step-up dosing in the outpatient setting, such as strategies to reduce the risk of cytokine release syndrome (CRS) and the potential use of wearable technology and remote monitoring.

  • How to manage toxicities associated with bispecific antibody therapy for MM in community practice
    2026/09/15

The Multiple Myeloma Hub spoke with María-Victoria Mateos, University Hospital of Salamanca, ES. We asked about the management of toxicities associated with bispecific antibody therapy for multiple myeloma (MM) in community practice.

In this interview, Mateos discusses practical considerations for the management of adverse events (AEs) associated with bispecific antibodies in MM. Mateos outlines how to recognize and manage cytokine release syndrome (CRS) and immune effector cell-associated neurotoxicity syndrome (ICANS), as well as strategies to mitigate infection risk. Mateos also discusses the management of skin, nail, and oral toxicities associated with G-protein coupled receptor family C group 5 member D (GPRC5D)-directed bispecific antibodies.

  • How to transition patients to community-based bispecific antibody therapy
    2026/09/11

The Multiple Myeloma Hub spoke with Shaji Kumar, Mayo Clinic, Rochester, US. We asked about transitioning patients to community-based bispecific antibody therapy.

In this interview, Kumar discusses practical considerations for transitioning bispecific antibody therapy to community-based practice in patients with multiple myeloma. Kumar outlines strategies for monitoring and managing treatment-related toxicities, including cytokine release syndrome, neurological toxicities, cytopenias, and infections. Kumar also discusses patient education, supportive care, and the evolving role of less frequent and subcutaneous bispecific antibody administration in facilitating community-based treatment.

  • Strategies for managing belantamab mafodotin-associated ocular toxicities
    2026/07/02

During the Multiple Myeloma Hub Steering Committee Meeting on May 12, 2026, key opinion leaders met to discuss strategies for managing belantamab mafodotin-associated ocular toxicities in patients with multiple myeloma (MM). The meeting featured a presentation by guest speaker Lisa Leypoldt, University Medical Center Hamburg-Eppendorf, followed by a faculty discussion on the practical challenges of monitoring, managing, and counseling patients receiving belantamab mafodotin-based therapies.

During their presentation, Leypoldt reviewed the mechanisms underlying belantamab mafodotin-associated ocular toxicity, highlighting the role of off-target uptake into corneal epithelial cells and the resulting transient visual symptoms. Leypoldt discussed the clinical manifestations of ocular events, including keratopathy, changes in visual acuity, and patient-reported symptoms, before outlining evidence-based approaches to monitoring and management. Leypoldt also reviewed data from the DREAMM-7 (NCT04246047) and DREAMM-8 (NCT04484623) studies, evaluating the role of dose modifications and extended dosing intervals in managing ocular toxicity. The subsequent faculty discussion focused on multidisciplinary care, patient counseling, and practical considerations for integrating ocular toxicity monitoring into routine clinical practice.
